    Refactoring for introducing and tuning parallelism for heterogeneous multicore machines in Erlang

EU COST Action IC1202: Timing Analysis On Code-Level (TACLe), and by a travel grant from EU HiPEAC.This paper presents semi‐automatic software refactorings to introduce and tune structured parallelism in sequential Erlang code, as well as to generate code for running computations on GPUs and possibly other accelerators. Our refactorings are based on the lapedo framework for programming heterogeneous multi‐core systems in Erlang. lapedo is based on the PaRTE refactoring tool and also contains (1) a set of hybrid skeletons that target both CPU and GPU processors, (2) novel refactorings for introducing and tuning parallelism, and (3) a tool to generate the GPU offloading and scheduling code in Erlang, which is used as a component of hybrid skeletons. We demonstrate, on four realistic use‐case applications, that we are able to refactor sequential code and produce heterogeneous parallel versions that can achieve significant and scalable speedups of up to 220 over the original sequential Erlang program on a 24‐core machine with a GPU.PostprintPeer reviewe

    Wake response to an ocean-feedback mechanism: Madeira Island case study

    This discussion focused on the numerical study of a wake episode. The Weather Research and Forecasting model was used in a downscale mode. The current literature focuses the discussion on the adiabatic dynamics of atmospheric wakes. Changes in mountain height and consequently on its relation to the atmospheric inversion layer should explain the shift in wake regimes: from a 'strong-wake' to a 'weak-wake' scenario. Nevertheless, changes in SST variability can also induce similar regime shifts. Increase in evaporation, contributes to increase convection and thus to an uplift of the stratified atmospheric layer, above the critical height, with subsequent internal gravity wave activity.Comment: Under review proces

This leads to humans becoming highly dependent on digital devices, including upon digital: assistance, intelligence, and decisions. A major concern of this digital dependence is the lack of human oversight or intervention in many of the ways humans use this technology. This dependence and reliance on digital technology raises concerns in how humans trust such systems, and how to ensure digital technology behaves appropriately. This works considers recent developments and projects that combine digital technology and artificial intelligence with human society. The focus is on critical scenarios where failure of digital technology can lead to significant harm or even death. We explore how to build trust for users of digital technology in such scenarios and considering many different challenges for digital technology. The approaches applied and proposed here address user trust along many dimensions and aim to build collaborative and empowering use of digital technologies in critical aspects of human society

    A Hybrid Approach to Parallel Pattern Discovery in C++

However, discovering places in sequential code where parallel patterns should be introduced is still highly non-trivial, often requiring expert manual analysis and profiling. We present a hybrid discovery technique to detect instances of parallel patterns in sequential code. This employs both static and dynamic trace-based analysis, together with hotspot detection.     Bridging topological and functional information in protein interaction networks by short loops profiling

    Protein-protein interaction networks (PPINs) have been employed to identify potential novel interconnections between proteins as well as crucial cellular functions. In this study we identify fundamental principles of PPIN topologies by analysing network motifs of short loops, which are small cyclic interactions of between 3 and 6 proteins. We compared 30 PPINs with corresponding randomised null models and examined the occurrence of common biological functions in loops extracted from a cross-validated high-confidence dataset of 622 human protein complexes. We demonstrate that loops are an intrinsic feature of PPINs and that specific cell functions are predominantly performed by loops of different lengths. Topologically, we find that loops are strongly related to the accuracy of PPINs and define a core of interactions with high resilience. The identification of this core and the analysis of loop composition are promising tools to assess PPIN quality and to uncover possible biases from experimental detection methods. More than 96% of loops share at least one biological function, with enrichment of cellular functions related to mRNA metabolic processing and the cell cycle.     Fluorofluorophores: Fluorescent Fluorous Chemical Tools Spanning the Visible Spectrum

    β€œFluoro” refers to both fluorescent and fluorinated compounds. Despite the shared prefix, there are very few fluorescent molecules that are soluble in perfluorinated solvents. This paucity is surprising, given that optical microscopy is a ubiquitous technique throughout the physical sciences and the orthogonality of fluorous materials is a commonly exploited strategy in synthetic chemistry, materials science, and chemical biology. We have addressed this shortage by synthesizing a panel of β€œfluorofluorophores,” fluorescent molecules containing high weight percent fluorine with optical properties spanning the visible spectrum.     Expression and function of Ξ±Ξ²1 integrins in pancretic beta (INS-1) cells

    Integrin-extracellular matrix interactions are important determinants of beta cell behaviours. The Ξ²1 integrin is a well-known regulator of beta cell activities; however, little is known of its associated Ξ± subunits. In the present study, Ξ±Ξ²1 integrin expression was examined in the rat insulinoma cell line (INS-1) to identify their role in beta cell survival and function. Seven Ξ± subunits associated with Ξ²1 integrin were identified, including Ξ±1-6 and Ξ±V. Among these heterodimers, Ξ±3Ξ²1 was most highly expressed. Common ligands for the Ξ±3Ξ²1 integrin, including fibronectin, laminin, collagen I and collagen IV were tested to identify the most suitable matrix for INS-1 cell proliferation and function. Cells exposed to collagen I and IV demonstrated significant increases in adhesion, spreading, cell viability, proliferation, and FAK phosphorylation when compared to cells cultured on fibronectin, laminin and controls. Integrin-dependent attachment also had a beneficial effect on beta cell function, increasing Pdx-1 and insulin gene and protein expression on collagens I and IV, in parallel with increased basal insulin release and enhanced insulin secretion upon high glucose challenge. Furthermore, functional blockade of Ξ±3Ξ²1 integrin decreased cell adhesion, spreading and viability on both collagens and reduced Pdx-1 and insulin expression, indicating that its interactions with collagen matrices are important for beta cell survival and function. These results demonstrate that specific Ξ±Ξ²1 integrin-ECM interactions are critical regulators of INS-1 beta cell survival and function and will be important in designing optimal conditions for cell-based therapies for diabetes treatment

    Young, healthy males and females present cardiometabolic protection against the detrimental effects of a 7-day high-fat high-calorie diet

    Purpose: High-fat, high-calorie (HFHC) diets have been used as a model to investigate lipid-induced insulin resistance. Short-term HFHC diets reduce insulin sensitivity in young healthy males, but to date, no study has directly compared males and females to elucidate sex-specific differences in the effects of a HFHC diet on functional metabolic and cardiovascular outcomes. Methods: Eleven males (24 ± 4 years; BMI 23 ± 2 kg.mβˆ’2; VΜ‡O2 peak 62.3 ± 8.7 ml.minβˆ’1.kgβˆ’1FFM) were matched to 10 females (25 ± 4 years; BMI 23 ± 2 kg.mβˆ’2; VΜ‡O2 peak 58.2 ± 8.2 ml.minβˆ’1.kgβˆ’1FFM). Insulin sensitivity, measured via oral glucose tolerance test, metabolic flexibility, arterial stiffness, body composition and blood lipids and liver enzymes were measured before and after 7 days of a high-fat (65% energy) high-calorie (+ 50% kcal) diet. Results: The HFHC diet did not change measures of insulin sensitivity, metabolic flexibility or arterial stiffness in either sex. There was a trend towards increased total body fat mass (kg) after the HFHC diet (+ 1.8% and + 2.3% for males and females, respectively; P = 0.056). In contrast to females, males had a significant increase in trunk to leg fat mass ratio (+ 5.1%; P = 0.005). Conclusion: Lean, healthy young males and females appear to be protected from the negative cardio-metabolic effects of a 7-day HFHC diet. Future research should use a prolonged positive energy balance achieved via increased energy intake and reduced energy expenditure to exacerbate negative metabolic and cardiovascular functional outcomes to determine whether sex-specific differences exist under more metabolically challenging conditions

    Polymeric Micelles in Anticancer Therapy: Targeting, Imaging and Triggered Release

    Micelles are colloidal particles with a size around 5–100Β nm which are currently under investigation as carriers for hydrophobic drugs in anticancer therapy. Currently, five micellar formulations for anticancer therapy are under clinical evaluation, of which Genexol-PM has been FDA approved for use in patients with breast cancer. Micelle-based drug delivery, however, can be improved in different ways. Targeting ligands can be attached to the micelles which specifically recognize and bind to receptors overexpressed in tumor cells, and chelation or incorporation of imaging moieties enables tracking micelles in vivo for biodistribution studies. Moreover, pH-, thermo-, ultrasound-, or light-sensitive block copolymers allow for controlled micelle dissociation and triggered drug release. The combination of these approaches will further improve specificity and efficacy of micelle-based drug delivery and brings the development of a β€˜magic bullet’ a major step forward
